Itsy Bitsy Stike Craft

I'm trying to make a strike craft that is no larger than 250 tons, has stealth capabilities and can hold at least one missile in a box launcher.

Similarly I am trying to make a missile that is highly damaging and super fast.

The trick is to make a strike craft and missile that can close in without being detected and launch a missile that hits within 5 seconds.

If this is indeed possible at what tech level does it become feasible? How big should the missile be? Should I opt for more box launchers or bigger missiles?

I can't see it being possible. Any decent opponent will have an anti-missile sensor that can detect missiles from more than 5 seconds off, and no amount of stealth on a fighter will protect you from being detected on a resolution 1 sensor.
