NPR neighbor with whom I am at war was tagged as hostile, but not this first contact raider.

His ship has just been blowed in few seconds.

Can ECM or cloaking interfere with STO fire ability ? Even if the ship is at zero distance from the colony, no fire occurs.

The answer to your question over Cloak and ECM is yes. Usually, they shorten your sensor view forcing you to shoot a closer range, not a 0 distance though. Besides, I am also assuming the target your are trying to shoot down has been properly marked as hostile. I am saying just because it did happen to me a few times that, especially on first encounters, due to the neutrality of the contact assigned on the diplomacy screen, I was not able to coerce my crew in shooting anyway and leave the diplomatic chit chats to the politicians after the threat was neutralized.  ;)

I just had my first fight using STOs defensively in v2.1.1. I had others in previous versions, and I never encountered a problem, but this time i am unable to get them to shoot their target. not sure if anything changed in the latest version, if it's a bug, or if (more likely) I forgot something :)

On a colony attacked by raiders, I have 5 formations consisting of 2 STOs units each :

None open fire on the ship, while it is within range of fire control.

Active sensors are ON for the 5 formations, and I also tested to SM a ship with a classic active sensor.

I tried to change STO targeting to all possibilities, WPT set to 0 or 1.

SM new STO templates on the colony, they don't open fire either.

If I SM a warship on the colony, combat takes place normally.

What I know about the attacking ship :
